Platele 3-phos patholo Barnes Kato e cellula IP3, via 1987; Snyder, 1989). Modulatory activities linked to intracellular receptors have also been suggested, based on differential effects of PAF antagonists, but they are less well characterized (Stewart et al., 1990; Hwang, 1990; Marcheselli and Bazan, 1994; Kato et al., 1994). PAF has been found also in lower eukaryotes, such as Tetrahymena pyriformis (Lekka et al., 1990) and Dictyostelium discoideum (Bussolino et al., 1991). D. discoideum cells grow as free-living amoebae, by feeding on bacteria. Development is triggered by starvation, and results in the formation of multicellular aggregates which undergo a series of morphogenetic changes (Bonner, 1967; Loomis, 1982). A major regulatory role in aggregation is played by cAMP, which is secreted by the cells a few hours after starvation and acts as a chemoattractant. In Dictyostelium, the intracellular concentration of PAF increases during development, and in response to cAMP (Bussolino et al., 1991). Spontaneous, periodic cellular oscilcur s, 19 n of leve lar et itan e ab s n guanylate cyclase. In contrast, PAF induces extracellular Ca2+ influx, independently of the presence of endogenous or exogenously added cAMP (Sordano et al., 1993). We have further characterized this effect of PAF, and we show that a specific, intracellular acceptor for PAF exists in these cells that appears during spike-shaped oscillations.

P30: Effects of Hemin on Ca2+Influx in Neurons of C57BL/6 Mouse Brain
Excitotoxicity results in a significant increase in Ca2+ influx; essentially from open N-Methyl-D-aspartate receptors (NMDARs) channels that cause a secondary rise in the intracellular Ca2+ concentration. It is correlated with neuronal death induced by Ca2+ overload. Dysfunction of NMDARs is associated with excitotoxic neuronal death in neurodegenerative disorders. In this study, the effects of...
متن کاملThe kinetics of changes in intracellular calcium concentration in fura-2-loaded human platelets.
We have investigated the sub-second kinetics of changes in cytosolic free calcium, [Ca2+]i, in fura-2-loaded human platelets by stopped-flow fluorimetry. Thrombin, vasopressin, platelet-activating factor, and the thromboxane A2 analogue U46619 all evoked a rise in [Ca2+]i which was delayed in onset by 200-400 ms in the presence of 1 mM external Ca2+. The responses to these agonists in media con...
متن کاملThe contractile action of platelet-activating factor on gallbladder smooth muscle.
Platelet-activating factor (PAF) may be a mediator of some sequelae of cholecystitis, a disorder with gallbladder motor dysfunction. The aims of this study were to determine the effect and mechanism of PAF on gallbladder muscle. Exogenous administration of PAF-16 or PAF-18 caused dose-dependent contractions of gallbladder muscle strips in vitro with threshold doses of 1 ng/ml and 10 ng/ml, resp...
متن کاملProstaglandins, Histamine and Platelet Activating Factor: Different Mediators in Dithranol-Induced Skin Damage
Dithranol is a potent agent in treating psoriasis but its adverse effects on intact skin have limited its usage. There are many proposed mediators for its adverse effects including prostaglandins, histamine, platelet activating factor and free radicals. In this study we examined the effect of different agents (diazepam, terfenadine, indomethacin and garlic extract) on dithranol-induced skin dam...
متن کاملCalcium influx: an intracellular message of the mitogenic action of insulin-like growth factor-I.
When G0-arrested BALB/c 3T3 cells were treated sequentially with platelet-derived growth factor and epidermal growth factor, cells became responsive to insulin-like growth factor-I (IGF-I). In these primed competent cells, 1 nM IGF-I elicited an approximately 3-fold increase in the calcium influx rate. IGF-I-induced calcium influx was relatively slow in onset and continued for at least 2 h in t...
